Estudio de compatibilidad G0/excipientes mediante métodos isotérmicos y no isotérmicos

El propósito del presente trabajo fue evaluar la compatibilidad del 2-(2-nitrovinil) furano (G-0), ingrediente farmacéutico activo (IFA) con actividad farmacológica como antiprotozoario en fase preclínica, con excipientes farmacéuticos de uso común en formas sólidas (Lactopress, Manitol, Dextrosa, Avicel PH102, Almidón de trigo, Sacarosa, Cloruro de sodio y DI-CAFOS), así como con excipientes empleados como agentes solubilizantes y estabilizantes, como la Dimetil-beta-ciclodextrina, Hidroxipropil-beta-ciclodextrina, Sulfobutil éter-beta-ciclodextrina y Sílica mesoporosa ordenada (SBA-15). La Calorimetría Diferencial de Barrido fue utilizada primeramente para detectar de forma rápida variaciones en las temperaturas de los picos y/o la entalpía asociada a diez mezclas físicas IFA/excipiente (1:1), que pudieran indicar signos de interacción. Adicionalmente se sometieron las mezclas a estrés isotérmico con el objetivo de acelerar las posibles interacciones y detectar incompatibilidades en un corto plazo. Las muestras envejecidas se analizaron mediante Cromatografía Líquida de Alta Eficacia (CLAE), Espectroscopía Infrarroja con Transformada de Fourier (FT-IR) y nuevamente se evaluaron por DSC. Las mezclas también fueron evaluadas desde el punto de vista organoléptico. El empleo de estas técnicas permitió confirmar la incompatibilidad del G-0 con Sílica Mesoporosa ordenada (SBA-15), DI-CAFOS y Avicel PH102, los cuales deberán evitarse en el desarrollo de futuras formulaciones del fármaco y la posibilidad de incluir con seguridad en ellas a las tres ciclodextrinas evaluadas.
The aim of the present work was to evaluate the compatibility of 2-(2-nitrovinyl) furan (G-0), an Active Pharmaceutical Ingredient (API) with pharmacological activity as antiprotozoal agent in preclinical phase, with pharmaceutical excipients of common use in solid dosage forms (Lactopress, Mannitol, Dextrose, Avicel PH102, Wheat Starch, Saccharose, Sodium chloride, and DI-CAFOS), as well as with excipients used as solubility and stability agents, such as Dimethyl-beta-cyclodextrin, Hydroxypropyl-beta-cyclodextrin, Sulfobutyl ether-beta-cyclodextrin and Ordered Mesoporous Silica (SBA-15). Differential Scanning Calorimetry was used firstly to detect variations in the temperature of the picks and/or the enthalpies associated for ten physical mixtures API/excipient (1:1) that could indicate signals of interaction. Additionally the samples were subjected to an Isothermal Stress Testing with the objective to accelerate the possible interactions and detect incompatibilities in a short term. The aged samples were analyzed by High Performance Liquid Chromatography (HPLC), Infrared Spectroscopy with Fourier Transformed (FT-IR), and then they were evaluated again by DSC. Additionally, the mixtures were evaluated by visual inspection. The use of these techniques allowed to confirm the incompatibility of G-0 with Ordered Mesoporous Silica (SBA-15), DI-CAFOS and Avicel PH 102, which should be avoided in the future formulations of the API and the possibility to include in them the three cyclodextins evaluated.
