Potencial genotóxico de Agave brittoniana Trel. subespecie brachypus mediante el ensayo de micronúcleos transplacentarios

Resumen

Se determinó el potencial genotóxico en el ensayo de micronúcleos transplacentarios de un crudo de saponinas de Agave brittoniana subespecie brachypus, especie que se le reconocen un conjunto de metabolitos que le confieren actividad antiparasitaria, antiinflamatoria y antifúngica. Se diseñaron cinco grupos experimentales de madres gestantes de la línea BALB/c/Cenp., un control positivo con ciclofosfamida (50 mg/Kg), un control negativo con agua destilada (10 ml/Kg) y tres grupos con los crudos de prueba a las dosis de 50, 100 y 200 mg/Kg, los cuales fueron tratados los días 14, 15 y 16 de la gestación. Al efectuar la cesárea fueron tomados cinco fetos por madre los que constituyeron los grupos del seis al 10 de la investigación. Fueron determinados la frecuencia de eritrocitos micronucleados, los índices de genotoxicidad y citotoxicidad en la médula ósea de las madres y el hígado de los fetos, mientras que en la sangre periférica de ambos solo se determinó el índice de genotoxicidad mediante la presencia de reticulocitos micronucleados. La sustancia en estudio no mostró genotoxicidad ni citotoxicidad, no aumentó significativamente la frecuencia de células con micronúcleos en médula ósea de las madres ni en hígado fetal, ni la frecuencia de reticulocitos micronucleados en sangre periférica (p≤0,05). Aun así, el crudo a 200 mg/Kg evidenció una tendencia hacia los valores del control positivo. Los resultados demostraron que el crudo de saponinas de Agave brittoniana subespecie brachypus no es genotóxico en el ensayo aplicado según las dosis evaluadas y el modelo seleccionado.
The genotoxic potential of Agave brittoniana subsp. brachypus, saponin crude was determined in the test of transplacental micronucleus. This species is recognized a set of metabolites that give anti-parasitic, anti-inflammatory and antifungal activity. Mothers' mice pregnant of the line BALB/c/Cenp. were distributed in five experimental groups, a positive control with cyclophosphamide (50 mg/Kg), a negative control with distilled water (10 ml/Kg) and three groups treated with the crude under evaluation (50, 100 y 200 mg/Kg), which were treated the days 14, 15 and 16 of the gestation. Five fetuses were taken by mother after caesarean. These fetuses constituted the groups from the six to ten of the research. The frequency of micronucleated bone marrow cells, the genotoxicity and citotoxic indexes were determined in the bone marrow of the mothers and the liver of the fetuses, while in peripheral blood alone the genotoxicity index was determined by presence of micronucleated reticulocytes. The saponin crude of Agave didn't show genotoxicity neither citotoxic activity. It didn't increase the frequency of cells significantly with micronucleus in the mothers' bone marrow neither in fetal liver, the frequency of micronucleated reticulocytes in peripheral blood (p≤0,05). Even so, a trend to the values of positive control in saponin crude to 200mg/kg was observed. The results demonstrated that the crude of saponins of Agave brittoniana subsp. brachypus is not genotoxic in the test applied according to the evaluated doses and the selected model.
