Resumen
En la presente investigación se evaluaron los efectos de algunos factores químico-físicos sobre la formación del complejo azul de molibdeno con vistas a la optimización del conocido método Actividad Antioxidante Total, para la estimación del potencial antioxidante de productos de origen natural. Mediante la aplicación de un Diseño Factorial (34) se comprobó la gran influencia de la concentración de ácido y la temperatura en la formación del complejo de polioxometalato, resultando el tipo de ácido y el tiempo de incubación pocos significativos. El ácido ascórbico mostró un menor tiempo de desarrollo del color en la formación del complejo que los metabolitos de referencia empleados. El empleo de cosolventes (etanol) influyó negativamente en la cantidad de complejo formado. Debido a lo anterior las condiciones propuestas para desarrollar el método fueron: Tipo de ácido: HCl; concentración de ácido/pH de la mezcla reactiva, 0.01 N, ~ 3; temperatura de incubación: 65 ºC, y tiempo de incubación: 40 min. Bajo estas condiciones experimentales el orden de potencia antioxidante para los productos evaluados fue el siguiente: pirogalol, quercetina, ácido ascórbico, ácido gálico y rutina.
In the present investigation, evaluate the effects of some chemical-physical factors on the formation of the blue molybdenum complex with a view to optimizing the known Total Antioxidant Activity method, for the synthesis of the antioxidant potential of products of natural origin. By applying a Factorial Design (34) the great influence of the acid concentration and temperature on the formation of the polyoxometalate complex is verified, resulting in the type of acid and the incubation time few contaminants. Ascorbic acid showed a shorter color development time in complex formation than the reference metabolites used. The use of cosolvents (ethanol) negatively influenced the amount of complex formed. Due to the above, the conditions proposed to develop the method were: Type of acid: HCl; acid concentration / pH of the reaction mixture, 0.01 N, ~ 3; incubation temperature: 65 °C, and incubation time: 40 min. Under these experimental conditions, the order of antioxidant potency for the products evaluated was as follows: pyrogallol, quercetin, ascorbic acid, gallic acid and rutin.
